Pliskova and her sister started training once a week under a coach in their home town of Louny, at the tender age of 4. She turned pro in 2009, and began her WTA career in 2012 playing her first senior Grand Slam tournament at the French Open, but lost in the first round to Marion Bartoli in the straight sets. In 2013, after exiting the Brisbane International and Sydney Open in the first round, she won her first WTA title at the Malaysian Open defeating the American player Bethanie Mattek-Sands in three sets.

 The same year, she with her twin sister Kristyna won the WTA doubles title at the General Ladies Linz to become the first set of twins in WTA history to win a double title together on tour. 2014 was a good year for Pliskova, having reached the semifinals and finals in a couple of major tournaments, she broke into the top 50 rankings and later kept performing so well that she ended up climbing up to world rank 24 after defeating Varvara Lepchenko in Hong Kong to get her second career title.

Despite losing several finals, Pliskova ended number 11th in the rankings in 2015 and also became the 13th most popular player of the year according to WTA's website, but not before she won her first Fed Cup title as a doubles team. She began 2016 by representing Czech Republic in the Hopman Cup partnering with Jiri Vesely, and has recorded singles victory against Jarmila Wolfe and Victoria Duval.
